Abstract

Flow cytometric measurements during germination of Primula vulgaris Huds. seeds.

The cell cycle activity was studied by FCM in five germinating cultivars of Primula vulgaris seeds within 12 days afterr the salt of imbibition in a 0.2% KNO3 , solution. The DNA replication can be quickly analysed with total seeds. Only 10 seeds per replication were necessary. At the beginning of germination (dry seeds) most of the cells were arrested in GI/GO of the cell cycle, in line with 2C DNA levels. Only O.41% to 1.0% of cells were arrested with 4C DNA levels. The percentage of endosperm nuclei (3C + 6C) was 62% to 71% of the total numbel of nuclei. The endosperm nuclei declined rapidly after radicle protrusion. The percentage of G2 nuclei increased shortly before the radicle protrusion started and after radicle emergence. The rate of DNA replication differs between the seed batches in relation to the seed quality (protrusion rate). Ten days after imbibition 0% to 98%ot the seeds had developed a radicle. The DNA replication mainly starts in the cotyledon and radicle region but also in the hypocotyls.
